This is a form of gelatin based chromatography which relies on an electric field to separate fragments of DNA, based on the size of the fragment.





DNA is extracted from an organism and then ';amplified'; by using special enzymes to manufacture billions of copies of the DNA. Next a special enzyme is introduced which splits all the DNA according to a particular base pair. This cleaves the DNA into fragments based on where certain pairs exist along the chain. The result is a mixture of fragment sizes whose sizes are unique to the DNA originally extracted. It is stained, placed in a small depression in a gelatin plate and an electric current applied. The current will then carry the fragments through the gelatine according to the size of the particles. The result is a series of bands, precisely positioned for a particular sample. If an unknown sample is analyzed in this manner, the DNA will be an exact match if the resulting pattern matches the original. This is because the DNA sequence will have been exactly the same, and hence the fragmentation pattern will also be identical. Because DNA is a hereditary material, kinship can be deduced by how closely the gelatine band pattern matches between two samples. The closer the match, the closer the two doners are related to one another. For nearly all multicellular organisms on earth, the match is at least 50% identical.
